MS 5068 comprises correspondence, diaries (1934-1951), reports, notes, texts of speeches, printed material and newspaper cutting books (1933-1972). The material relates to his work for the Australian Aborigines, and his involvement in various organizations, such as the Aborigines Advancement League of South Australia. It documents the development of the Ernabella Mission in the Musgrave Ranges, in 1937, and the use made of the Smith of Dunesk Bequest for the improvement of Aboriginal welfare. The bulk of the collection covers the period from 1930 to 1960 when Duguid was campaigning most actively for Aboriginal rights. The later correspondence, from 1960 to the early 1980s, consists mainly of congratulatory notes and telegrams on Duguid's 80th and 90th birthdays, and on his award of the O.B.E. The collection also includes the typescript of his autobiography (11 boxes).
MS Acc03.202 comprises a box of medals won by Dr Charles Duguid. They include athletic awards as well as Imperial honours (1 box).
MS Acc03.292 contains correspondence, newspaper cuttings, journal articles, photographs, albums, printed articles and monographs, souvenirs and sketch maps relating to Duguid's work with the Aboriginal community at the Ernabella Mission. Also includes two albums of photographs from his service in Egypt and Palestine in World War 1, one of which includes an image of aviator Ross Smith, as well as papers and notes on his training as a medical practioner. A significant negative collection of approximately 700 images is also contained in the papers and likely represents the contents of the photograph albums. Items and articles of anthropological and linguistic interest are also included (20 boxes, 1 archive box, 4 large folio boxes, 2 small folio boxes, 3 folio items).

Duguid moved to Australia in 1912 and became a practising gynaecologist and obstetrician in Adelaide. From 1928 he was actively involved in organisations such as the Aborigines' Advancement League of South Australia, the Council for Aboriginal Rights and the Association for the Protection of Native Races. In 1970 Duguid was awarded the O.B.E. for his work with Aborigines, and in 1974 received the Anisfield-Wolf Award in Race Relations for his autobiography "Doctor and the Aborigines".
